Intended use:
For in vitro diagnostic use. This RIDA®FLUOR Legionella IgG tests are immunofluorescence assays (IFA) for qualitative detection of IgG antibodies against Legionella pneumophila serogroup 1 – 14 and six non-pneumophila-species of Legionella (L. bozmanii, L. dumoffi, L. gormani, L. jordanis, L. longbeachae, L. micdadei) in human serum. The test should be used in reasonable suspected cases of Legionella infections or for determination of an immune status.
General information:
Standard tests for antibody detection (IFA, ELISA) have been established as laboratory tests, since cultural growth of the organism from BAL (broncho-alveolar-lavage) samples is difficult. We provide under the trademark RIDA®FLUOR one variation with three pools of our IFA, which use Legionella pneumophila and non-pneumophila-species as antigen.
Test principle:
Inactivated Legionella pneumophila- and non-pneumophila-species are fixed as different pools (pool I – III) in defined wells on the surface of test slides. Diluted serum samples are given to the wells. Present anti-Legionella-antibodies bind to cell surfaces and form antigen-antibody-complexes. After a washing step for removing unbound antibodies, the complexes become visible by adding FITC-conjugated anti-human-IgG-immunoglobuline. After a further washing step for removing excess conjugate, the test can be analyzed by a fluorescence microscope with 400x magnification.
