The RIDA®LINE Helicobacter tests are line blot for specific detection of IgA or IgG antibodies against Helicobacter pylori in human serum. The tests should be used for confirmation of positive results or clarification of unspecific findings with other serological methods.
By using type-specific antigens it is also possible to differentiate between an infection with type I and type II.
The diagnostic strategies for H. pylori can be divided into invasive and non-invasive methods. With the invasive gastroscopy a histological investigation of the derived biopsy material is combined with the detection of organism’s urease activity in the biopsy material. But, the biopsy can lead to false negative results, since material can be removed from places of the stomach where Helicobacter pylori has not colonized. With the identification of serum antibodies or the detection of H. pylori antigen in stool the non-invasive diagnosis by enzyme immunoassay gains increasing importance in view of its advantages for the patients. Herewith, the enzyme immunoassay is distinguished by its easy handling as a screening method. Additionally, the line blot has the advantage that it differentiates between type I and II H. pylori. Herewith, an analysis of the different Helicobacter pylori types with their clinical significance is made possible for epidemiological investigations.
For this line immunoassay, highly purified recombinant antigens of Helicobacter pylori are fixed on nitrocellulose membrane strips. These strips will be incubated with the samples (serum, plasma). Specific antibodies present in the samples will bind to the pathogen antigens on the strip. After removing the unbound antibodies by washing, a second incubation with anti-human IgA or IgG conjugate follows. In the last step (staining reaction), specifically bound antibodies against the different Helicobacter pylori antigens become visible as dark bands on the strips.
